Despite recent improvements, treatment options for hepatocellular carcinoma (HCC) remain largely unsatisfactory. Epidrugs are currently being evaluated as cancer therapy and a first inhibitor of the histone methyltransferase Enhancer of zeste homolog 2 (Ezh2) recently obtained FDA approval. Increased expression of the epigenetic writer Ezh2, responsible for H3K27 di- and trimethylation (H3K27me2 and H3K27me3) within Polycomb repressive complex 2 (PRC2), is frequently detected in HCC tissues and correlates with the aggressiveness and/or poor prognosis of HCC. Ezh2 knockdown in HCC cells reverses tumorigenicity in a nude mouse model, suggesting a potential therapeutic value of Ezh2 inhibition in HCC. Ezh2 activity is regulated by post-translational modifications, including glycosylation by O-linked N-acetylglucosamine (O-GlcNAc) transferase (OGT). Interestingly, it has be reported that OGT and Ezh2 co-repress a subset of potential tumor suppressor genes in breast cancer MCF-7 cells.
